I'm transferring from a school that didn't have no Greek life so I really want to join but I'm going to be a junior in the fall. Will my age work against me when it comes to joining a fraternity? I'll also be 21!

I'm in the same situation, highly considering transfer.
Also, are upperclassmen allowed to rush fall?

Can't hurt to try. Freshmen can't rush in the fall but everyone else can which can work to your advantage but also the pledge classes are smaller. The fact that you're 21 might help if you look like a brother for ID purposes.
