do sororities actually check through the school to see your gpa?

Yep, when you register for recruitment I think you have to give your GPA and then everything's updated when the semester's over. They need to check to make sure you meet whatever requirement they have.

Is it based on first semester gpa or gpa at the end of the year?

It's fall and includes winter if you take it

Also, the greek life office can just look it up themselves as well. Doesn't matter if you give it or not.
