I wouldn't say it DEFINES you, but greek life is definitely huge at mizzou and fairly competitive at least for mid tier and top tier houses. But at the same time, not being involved in a sorority is by no means the end of the world. Mizzou is a huge school, you will find your niche and group of friends. You will still be able to go out just the same with the exception of date parties and mixers, which are honestly not a big deal. Overall, yes it's big. You will see girls in their letters everywhere. People do often ask "what house are you in?" as one of the first questions in a conversation because it's almost assumed sometimes. But like I said, not being in a sorority is a perfectly fine route to take too!
